Hydrogen peroxide is a part of the oxygen reduction pathway.  It is either produced by the two-electron reduction of molecular oxygen or by the one-electron reduction of superoxide anion radical. With hydrogen peroxide being a potent oxidant, the levels of hydrogen peroxide must be accurately determined in order to fully characterize the system’s oxidative state .

National Diagnostics Hydrogen Peroxide Assay Kit is a rapid, sensitive, and quantitative method that helps determine hydrogen peroxide in chemical or biological systems. The assay is based when a complex between xylenol orange and ferric iron forms, produced by the peroxide-dependent oxidation of ferrous iron. This reaction is quantified colorimetrically, detecting as little as 15ng/ml of peroxide. Linear results can be obtained up to approximately 1000ng/ml.

Protocol

Hydrogen Peroxide Assay Protocol

Assay Reagent Preparation

Prepare 1.8ml of reagent per sample to run tests in duplicate. To prepare 20 ml combine 19.8 ml of Component A with 0.2ml Component B. The mixture is stable at room temperature for one working day.

Assay Procedure

  1. Mix 0.9 ml of Assay Reagent with up to 0.1 ml of sample.
  2. Incubate at room temperature for at least 30 minutes to allow for complete color development.
  3. Read absorbance at 560nm.
